Sample Preparation
We suggest prior experimentation with clean (undiluted) samples, 1:2 or 1:4 dilutions. Avoid diluting your samples more than 1:10, as this will exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit.

Assay Principle: The ASCAb ELISA kit applies the competitive enzyme immunoassay technique using Saccharomyces cerevisiae antigen and an ASCAb-HRP conjugate. The test sample and buffer are incubated together with the ASCAb-HRP conjugate in a precoated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for the HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow.
Color intensity is measured spectrophotometrically at 450 nm in a microplate reader. Color intensity is inversely proportional to ASCAb concentration, as ASCAb in the samples and the ASCAb-HRP conjugate compete for the Saccharomyces cerevisiae antigen-binding site. Since the number of sites is limited, as ASCAb occupies more sites in the sample, fewer sites remain to bind the ASCAb-HRP conjugate. A standard curve is drawn that relates the colour intensity (D.O.) to the concentration of the standards. The concentration of ASCAP in each sample is interpolated from this standard curve.
