For research use only

For research use only. It should not be used in diagnostic procedures.

Species reactivity: Mouse

Samples

Serum, plasma, cell culture supernatants, body fluids, and tissue homogenates

Test type: sandwich

Detection range: 10-250u/mL

Intended uses

This ASCAP ELISA kit is a 1.5-hour solid-phase ELISA designed for the quantitative determination of mouse ASCAP. This ELISA kit is for research use only, not for therapeutic applications!

Preparation and Storage

Store all reagents at 2-8 degrees C.

Sample Preparation

We suggest prior experimentation with clean (undiluted) samples, 1:2 or 1:4 dilutions. Avoid diluting your samples more than 1:10, as this will exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit.

Product note

Information selected from the online data sheet is extracted from bioinformatics databases, sometimes resulting in the ambiguous or non-relevant product information. It is the customer’s responsibility to review, verify and evaluate the information to ensure it matches their requirements prior to purchasing the kit. Our ELISA Kit assays are dynamic research tools and may be updated and improved from time to time.

If the format of this assay is important to you, please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current trial details. We cannot guarantee that the sample manual posted online is the most up-to-date manual, it is intended to serve as an example only. Consult the instructions for use provided with the assay kit for precise details.

Other notes

Occasionally, small volumes of ASCA ELISA kit vials may become trapped in the product vial seal during shipping and storage. If necessary, briefly spin the vial in a tabletop centrifuge to dislodge any liquid in the vial cap. Certain products may require dry ice shipping and an additional dry ice fee may apply.

MBS733639 is a ready-to-use microwell and strip plate ELISA (Enzyme-Linked Immunosorbent Assay) Kit for testing the presence of anti-Saccharomyces cerevisiae HCP ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls generate a theoretical detection range of the kit in biological research samples containing ASCA.

The analytical biochemical ELISA technique of the MBS733639 kit is based on ASCA antibody-ASCA antigen (immunosorbent) interactions and an HRP colourimetric detection system to detect ASCA antigen targets in samples. The ELISA Kit is designed to detect native, non-recombinant ASCA. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, and secretions. Quality control assays that evaluated reproducibility identified intra-assay CV (%) and inter-assay CV (%).

Product related information for kit ELISA ASCA

Assay Principle: The ASCAb ELISA kit applies the competitive enzyme immunoassay technique using Saccharomyces cerevisiae antigen and an ASCAb-HRP conjugate. The test sample and buffer are incubated together with the ASCAb-HRP conjugate in a precoated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for the HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow.

Color intensity is measured spectrophotometrically at 450 nm in a microplate reader. Color intensity is inversely proportional to ASCAb concentration, as ASCAb in the samples and the ASCAb-HRP conjugate compete for the Saccharomyces cerevisiae antigen-binding site. Since the number of sites is limited, as ASCAb occupies more sites in the sample, fewer sites remain to bind the ASCAb-HRP conjugate. A standard curve is drawn that relates the colour intensity (D.O.) to the concentration of the standards. The concentration of ASCAP in each sample is interpolated from this standard curve.
